Development and Validation of Models for Alzheimer’s Disease-Related Dementias (ADRD) (R61/R33 – Clinical Trial Not Allowed)

While there are multiple models of ADRDs that capture some aspects of disease, more work needs to be done to fully model all aspects of these complex diseases, advance knowledge of newer or multiple pathologies, integrate vascular elements, and incorporate relevant clinical, pathologic, and molecular phenotypes. Novel cellular and animal models of multiple interacting pathologies, that mimic the multiple interacting risk factors and comorbidities seen in patients, and that advance knowledge of key molecular pathways in a pathophysiologically-relevant context are particularly needed.

This NOFO is intended to support the development of validated, clinically- and/or pathophysiologically-relevant models (this could include small animals, larger animals, ex vivo models, human iPSCs, organoids, tissue-on-chip, or other in vitro models). All models must have translatability to human studies. For the purposes of this NOFO, small animal models are defined as invertebrates and smaller vertebrates such as zebrafish and rodents. Large animal models would include, for example, dogs, pigs, and rabbits.
